Hereby, we would like to inform you that we have adapted our standard processes for the purification of antibodies via Protein G affinity chromatography. At InVivo, we are constantly striving to evaluate and further optimize existing processes in order to offer our customers the best possible product quality and highest yields. For purification via Protein G affinity Chromatography, InVivo has offered two different options so far, using either glycine/HCl or citric acid as elution buffer. Over the years, our internal tests have shown that elution via citric acid is by far the most advantageous option. This gentle elution at a weaker pH value induces less aggregate formation of the product and, at the same time, almost always leads to significantly higher product yields. In addition, the amine-free work ensures that the antibody solution can be used for a greater variety of diagnostic methods, since bioconjugation of the antibodies is possible without any problems. We have therefore decided to narrow down our set of standard purification methods and will routinely use elution in new projects via citric acid when purifying antibodies via Protein G affinity chromatography. Elution via glycine/HCl, on the other hand, can be used in the future on explicit customer request. This will take effect on January 2022. However, we would like to point out that products with fixed specifications are not affected by these modifications. Here, purification is carried out as specified by the customer.
